I have 2 SMTP services running on one machine. Why? because.  ;-)

     I have LSMTP listening to port 25 and another smtp service listening
     on port 600.

     I know to route all traffic to LISTSERV.DOMAIN.COM -> LISTSERV
     I want all other traffic that's going to DOMANI.COM to go to the smtp
     service on port 600.

     I've tried setting up the routing in LSMTP but I must not have it
     configured correctly.

     The routing I've tried so far are:
     DOMAIN.COM -> SMTP -> MyIPaddress:600
     *.DOMAIN.COM -> SMTP -> MyIPaddress:600

     Am I missing something?
     Do I need to turn mail relay on? If so how?
