LISTSERV Ver 14.3 has a new feature that permits cancelling the 48hr server-wide spam quarantine on specific addresses. This isn't a bypass or workaround, you (the Site Admin) actually cancel the quarantine for that address on that server. This of course restores the ability for that address to send messages. There is no way to do this on any earlier version of LISTSERV. For earlier versions, the 48 hr quarantine is absolute. (Manual editing of PERMVARS.FILE is absolutely out of the question, don't even think about it.) Note however that if that address again attempts to send an identical message to too many list addresses (or too many listname-request addresses) it will again be immediately caught by the spam trap. So, you also need to plan a way for this address to send the necessary messages without getting caught. List Owner lists, Super-lists, and other methods are the suggested routes to investigate.
