As of now, about an hour later, there are still 2,317 files in there. So apparently if these are files waiting to be queued and sent out, there must be something going on between the listserv and the transfer to the mail server?

The 3113 files should be less now if the exchange SMTP relay is interacting with the spool directory. If it is the same the relay is probably not connecting well to the listserv.

The queue viewer on Exchange 2010 is showing me 218 delayed items, about 90% of which are from the Listserv, both today's mailings and yesterday's.

Those are queued up for you SMTP relay I think. Which should be annotated in your server config settings.

I checked the SPOOL directory. Someone just sent out an email to the list at 11:37, and as of 12:09 there are 3,113 files in the SPOOL directory with .MAIL suffixes, all with today's date 2/7/2013.
I don't see an SMTPSVC directory. Is it on the Listserv server?

Also check that there are no issues with the SMTP process on the mail relay used by Listserv. If it is having problems with incoming connections that will cause mail to back in the Listserv outbound queue.
