For tracking down delivery issues, I love the SMTP header parser at http://mxtoolbox.com/EmailHeaders.aspx . It makes reading SMTP headers so much simpler and shows where the delay is very clearly. The biggest challenge is successfully instructing your irritated users to forward a message as an attachment to preserve the header.

I would seek the services of a 3rd party mass mailer who does this for a living.  We have both emergency and several informational email alerts for the different campuses which totals about 110K email subscribers as well as tweets and text messages for those who prefer those methods.
